Abstract: Successful generation and propagation of patient-derived xenograft models requires a supportive infrastructure composed of dedicated laboratory and animal facilities, regulatory approvals, detailed standard operative procedures, and, importantly, skilled technical staff.This chapter will detail the methodologies involved in the processing and implantation of freshly collected tumor samples and in the maintenance and propagation of successfully established PDX models. In addition, we will discuss the critical parameters influencing the likelihood of successful generation of a PDX and how to avoid the most likely pitfalls. © 2017 Elsevier Inc. All rights reserved.
