Post-transcriptional regulation of mRNA provides a rapid and powerful way to modulate the abundance of proteins in a cell. The multifunctional RNA binding proteins (RBP) mediate the various steps of RNA biogenesis. The riboproteome, comprised of RBPs bound to their target RNAs, provide an additional level of control that dictates cell fate, self-renewal, and cancer progression. Technological innovations have uncovered a role for RBPs in hematopoietic malignancies and a deeper understanding of their targets and function. This chapter describes a subset of RBPs that have dual functions in self-renewal and leukemia. Additionally, we examine how growth factor signaling pathways can alter their function and discuss the potential therapeutic strategies for targeting this novel class of proteins in the most aggressive cancers with stem-like properties. © 2014 by John Wiley & Sons, Inc.
