Article Title: | Tolerance of bone marrow cells in mice to high-dose arabinosylcytosine |
Abstract: | Aim: To investigate the feasibility of human cytidine deaminase gene (CD gene) transferred into mouse bone marrow cells and observe the tolerance of bone marrow cells to high-dose arabinosyleytosine (Ara-C). Methods: The study was conducted in the Central Laboratory of the First Affiliated Hospital of China Medical University from July 2003 to December 2004. Human cytidine deaminase gene was transferred into mouse bone marrow cells by means of co-culture with the retroviral vector. The co-cultured bone marrow cells and drug-treated Ara-C resistant colony-forming unit granulocyte-macrophage (CFU-GM) were observed. DNA was extracted from mouse bone marrow cells. The expression of drug resistance genes in mouse bone marrow cells after transfered by retroviral vector was determined by PCR. Results: The drug resistance colonies yield was 52% in bone marrow cells containing drug-resistance gene and 52 in the controls. Expression of CD gene in extracted DNA of bone marrow cells in transfected mice were dectected by PCR. The tolerance of bone marrow to Ara-C was remarkably increased after transfected by drug-resistance gene. Conclusion: CD gene can not only integrate and co-express in mice bone marrow cells, but also increase the drug resistance to Ara-C. |
