Abstract: In this chapter, we will firstly outline the discovery of Cerenkov luminescence and its physical principles to be followed by a discussion of the current state of the art along with best practices. The current range of preclinical and clinical implementations and developments will be highlighted including image-guided tumor resection, dosimetry monitoring and agent development. Future developments in this field, such as approaches to shift the blue weighted emission of CL into less scattered and absorbed red light will be discussed. © 2025 Elsevier B.V., All rights reserved.
