Abstract: This limiting dilution microculture assay is an in vitro quantitative assay for enumerating residual T lymphocytes in T cell-depleted BM. This assay requires only 1 x 106 BM cells. By determining the fraction of negative wells for each cell dose, one can determine the frequency of T lymphocytes in a given sample and thus calculate the absolute number of T lymphocytes in that sample. The sensitivity of the assay was such that 7.5 x 105 T lymphocytes were identified in a sample containing 1.6 x 109 mononuclear cells.
