Abstract: In vitro differentiation of mouse ES cells provides an efficient alternative assay for the study of genes that exhibit early lethality in vivo. SCL is a bHLH factor implicated in T-cell leukemia and known as the master gene of hematopoiesis. SCL-deficient mice die during gestation due to severe anemia and absence of primitive and definitive hematopoiesis. SCL is also expressed during brain development but its function in the CNS is unknown. We assayed the neural fate potential of ES cells exhibiting gain and loss of function mutations in SCL. Our in vitro differentiation system revealed unexpected dose-dependent negative regulation of midbrain dopamine neuron fates by SCL. Genome-wide differential expression screens identified potential downstream targets of SCL function in the CNS including Wnt1. Ectopic Wnt1 expression partially rescued dopaminergic differentiation of SCL transgenic ES cells implicating negative regulation of Wnt1 by SCL as a novel mechanism controlling midbrain development.
