Abstract: Immunotherapy is highly efficient for the treatment of mismatch-repair-deficient metastatic colorectal cancer. Two recent articles1,2 challenge the current paradigm of anti-PD-1 monotherapy for the frontline treatment of metastatic mismatch-repair-deficient colorectal cancer, as the combination of an anti-PD-1 and anti-CTLA-4 showed improved response rate and progression-free survival compared to chemotherapy or anti PD-1. © 2025 Elsevier Inc.
