This chapter discusses the histological preparation of pathology slides meant for whole slide imaging (WSI) and explains the additional quality control checks to minimize histological artifacts. The type of laboratory supplies, quality of reagents, tissue handling, and processing techniques, as well as other preanalytical variables at any point of slide preparation could introduce artifacts and ultimately affect the quality of the digitized images. In addition to the preanalytical and analytical factors that could influence the quality of scanned images, the WSI implementation process must evaluate the potential contributions of histological supplies such as types of tissue marking dyes used during grossing, glass slides, mounting medium, coverslip, and the quality of slide labels. This chapter provides complete overview of histological process as well as potential artifacts that could affect the image quality and recommends laboratories to validate their workflows with the newly implemented WSI devices. © 2025 Elsevier Inc.
